Abstract
This study evaluated the anti-inflammatory activity of the ethanol leaf extract of Pennisetum purpureum in cyclophosphamide-induced immunosuppressed Wistar rats using cyclooxygenase-2 (COX-2), nitric oxide (NO), prostaglandin E₂ (PGE₂), and C-reactive protein (CRP) as biochemical markers. Thirty-five male Wistar rats were randomly assigned into seven groups comprising a normal control, negative control, positive control, vehicle control, and three extracttreated groups (100, 200, and 400 mg/kg). Immunosuppression was induced with cyclophosphamide, while the extract was administered orally for 14 days. Serum COX-2, PGE₂, and CRP concentrations were determined using enzyme-linked immunosorbent assay (ELISA), whereas nitric oxide was estimated using the Griess reaction. Data were analyzed using one-way analysis of variance (ANOVA) followed by Tukey’s post hoc test at a significance level of p < 0.05. Cyclophosphamide administration significantly increased the levels of COX-2, NO, PGE₂, and CRP in the untreated group, confirming successful induction of inflammation. Treatment with Pennisetum purpureum leaf extract produced a significant dose-dependent reduction in all inflammatory markers, with the 400 mg/kg dose demonstrating effects comparable to the normal and standard drug-treated groups. The observed anti-inflammatory activity is attributed to the phytochemical constituents of the plant, particularly its flavonoids and phenolic compounds, which are known to modulate inflammatory pathways. The study concluded that the ethanol leaf extract of Pennisetum purpureum possesses significant anti-inflammatory activity and may serve as a promising natural source for the development of plant-based anti-inflammatory agents.
