Abstract
Natu’ral p’roducts ‘rich in phytochemicals have gained conside’rable attention fo’r thei’r antioxidant and p’rotective p’rope’rties. This study evaluated the antioxidant potential of the ethanol leaf ext’ract of Pennisetum pu’rpu’reum in cyclophosphamide-induced immunosupp’ression in Wista’r albino ‘rats th’rough phytochemical sc’reening, assessment of oxidative st’ress bioma’rke’rs, and histopathological examination of spleen tissues. Qualitative phytochemical analysis ‘revealed the p’resence of alkaloids, flavonoids, phenolic compounds, tannins, te’rpenoids, dite’rpenes, t’rite’rpenes, quinones, saponins, ca’rbohyd’rates, ca’rdiac glycosides and p’roteins/amino acids, while phlobatannins we’re not detected. Thi’rty-five Wista’r albino ‘rats we’re ‘randomly assigned into seven g’roups comp’rising a no’rmal cont’rol, negative cont’rol (induced but untreated), positive cont’rol (treatment with standard drug), vehicle cont’rol( treatment with sterile water) and th’ree ext’ract-t’reated g’roups. Oxidative st’ress bioma’rke’rs, including malondialdehyde (MDA), catalase (CAT), supe’roxide dismutase (SOD) and ‘reduced glutathione (GSH), we’re dete’rmined using standa’rd biochemical methods, while spleen tissues we’re p’rocessed fo’r histopathological evaluation using hematoxylin and eosin staining. The ‘results showed that t’reatment with the ext’ract significantly (p < 0.05) dec’reased MDA levels while significantly inc’reasing CAT, SOD and GSH activities in a dose-dependent manne’r. The high-dose g’roup (400 mg/kg) ‘resto’red antioxidant pa’ramete’rs to values compa’rable with the no’rmal cont’rol, whe’reas the vehicle cont’rol showed no significant imp’rovement. Histopathological examination ‘revealed nea’r-no’rmal splenic a’rchitectu’re in G’roups 3, 4 and 6, while G’roups 2, 5 and 7 showed mild pathological alte’rations, including focal nec’rosis, fib’rosis and pseudo’rosette fo’rmation, ‘respectively, indicating pa’rtial ‘recove’ry of splenic tissue. These findings suggest that the ethanol leaf ext’ract of Pennisetum pu’rpu’reum possesses significant antioxidant activity and p’rotects against cyclophosphamideinduced oxidative st’ress, p’robably due to its ‘rich phytochemical composition. The study suppo’rts the potential use of Pennisetum pu’rpu’reum as a natu’ral sou’rce of antioxidant compounds fo’r the management of oxidative st’ress-’related diso’rde’rs.
