Abstract

Millions of people around the world are widely consuming Tomato fruits in every season as it provides an easy and affordable source of nutrients to them. The study was conducted to estimate the microbial post-harvest spoilage of tomatoes from two different markets (Artisan and Ogbete) in Enugu State Nigeria. Different samples of tomatoes were collected from these Markets and were kept in a sterile bag where it was left open (for air-borne pathogens to decay them). Deterioration started on the 3rd day and samples were collected which after the 5th day of decay, more samples were collected, then the samples were used to; i. Isolate the microbe contaminants. ii. Determine the type of bacteria which contaminates the tomatoes plant. iii. Determine the important source of these contaminants. The samples were cultured onto MacConkey agar and Nutrient agar. The microbial quality was determined by quantifying the total viable cell count. The highest bacterial count (2.3x106 cfu/ ml) for tomato sample was found in sample D, collected from Ogbete Market and the lowest count was (1.0x105 cfu/ ml) found in sample F collected from Ogbete Market (Table 5). On the other hand, the highest total bacterial count (1.25x106 cfu/ ml) for tomato sample was found in sample F, Table 6 in Artisan Market, and the lowest count was observed to be 0.5 x103 cfu/ ml in another tomato sample A (Table 6). Gram stain reaction and biochemical proprieties were identified based on the chemical and physical properties of their cell wall. The isolated bacteria are; Gram-positive (Staphylococcus aureus, Bacillus cerus, Streptococcus pneumonaie) and Gram negative bacteria (Escherichia colistridium, Yersinia pestic,Proteus mirabilis). This study detected contaminants source of tomato spoilage during post-harvest storage and distribution.
